The term“coliform bacteria” doesn’t describe a taxonomical group beyond theeubacteria. It actually is more a summary of some genera from the family ofenterobacteria. Their common biochemical characteristics are positive lactosereaction as well as negative oxidase reaction. TheEnterobacteriaceae are gram-negative, rod-shaped bacteria which are typically1-5 µm in length. They are facultative anaerobes and most are motile, butnon-motile genera exist as well. P.aeruginosa is a rod shaped, gram-negative, aerobic bacterium which ismotile by unipolar flagella. Under anoxic conditions it is able to reducenitrate for energy generation. It has been identified as an opportunisticpathogen of both humans and plants and can cause a variety of diseases especiallyin people with weakened immune systems. P. aeruginosa and othermembers of the genus ( e.g. P. Putida ) are also known to occur as acause of spoilage in food products.

Duringincubation of the Compact Dry PA plate, the growing colonies will develop ared coloration which is due to the integrated redox indicator die Tetrazoliumsalt. TheGram-positive bacterium Bacillus cereus is able to produce longlasting spores and is commonly found in soil and water. Therefore, it entersthe food processing chain relatively easy. B. cereus can spoilfoods; furthermore, its emetic and diarrheagenic enterotoxins cause vomitingand diarrhea after consumption. Routine testing procedures for B.cereus are thus recommended.

Compact DryX-BC plates are incubated at 30° C for 24 hours. Due to the chromogenic substancesin the medium, B. cereus grows as green-blue colonies. If bacteriaother than Bacillus cereus grow on the plate, they usually producewhite colonies. Staphylococcusaureus is a ball shaped bacterium which can be observed as a grape-likeclustered organism under the microscope. Cells are sized between 0.8 and 1.2µm, Gram-positive and non-motile. S. aureus is a natural commensalof humans which lives on skin and upper airways and doesn’t cause diseases ingeneral. In case of infections with pathogenic strains or in immunodeficientpersons S. aureus is able to cause skin infections and abscessesbut also life threating diseases as pneumonia, meningitis, endocarditis, ortoxic shock syndrome ( TSS ).

One reason forthe facultative pathogenicity of S. aureus is its ability to formenterotoxins ( SET ), which can accumulate in contaminated food and can’t beinactivated totally by heat treatment. Contaminations of food stuffswith S. aureus are almost always caused by humans ( hands,sneezing, coughing ). S. aureus therefore is considered asindicator for poor personal hygiene. Beside its relevance for foodhygiene S. aureus also plays a significant role in hospitalhygiene. Generalinformation :

Standardmethods for determining the total viable bacterial count of heterotrophicbacteria in water usually recommend nutrient-rich media and incubation at 36± 1 °C. In some cases, only part of the actual microorganisms in the samplecan be detected under these conditions. Therefore, a longer incubation periodand a lower temperature are specified for the nutrient reduced Compact DryAQ. This ensures that stressed or chlorine tolerant bacteria can also berevitalized and cultivated. Microorganisms, such as bacteria, yeasts andmolds, can be cultivated using Compact Dry AQ.

The »ready-to-use«plate is also particularly suitable for the membrane filter method. A 47 mmmixed cellulose ester ( MCE ) filter with 0.45 μm pore size should be usedfor this task. After water filtration, the filter should be placed in themiddle of the previously moisturized Compact Dry plate completely free frombubbles and with the trap side facing upwards. After applying the watersample ( 1 ml ) or preparing saline solution ( 1 ml prior to filterapplication ), the dehydrated Compact Dry AQ plate becomes gel-like. Theliquid self diffuses evenly. Enterococciare Gram-positive organisms which belong to the intestinal bacteria as wellas the Gram-negative Enterobacteriaceae. Enterococci may appear ascontaminations in a variety of fermented foods. Their presence in foodproducts has long been considered as an indication of poor sanitaryconditions during production and processing. On the other hand, enterococciare specifically used as starter cultures for the fermentation processes of avariety of foods. It is claimed that enterococci play an important role inthe development of the organoleptic properties of the fermented foods. Forwater, the presence of enterococci serves as an indicator of fecalcontamination. Enterococci will only appear in water if they are inserted bycontaminations with human or animal feces.

Compact Dry SWAB NISSUI

Intended use :

Surfacesampling device consisting of a sterile swab in 1 ml sterile PBS/peptonewater buffer for sampling, transport and subsequent quantitative detection ofpresent microorganisms in combination with Compact Dry culture medium plates.

General information :

The CompactDry Swab is a tool for sampling surfaces in the production environment offood producers. The buffer within the tube is used to stabilize the collectedmicroorganisms during transport. The sample needs to be homogenouslydistributed in the buffer before applying it to a Compact Dry plate. To dropthe sample liquid onto the plate use the outlet in the bottom of the CompactDry Swab.

Inoculatedplates have to be incubated and evaluated according to the correspondingInstruction For Use which is given for each individual Compact Dry type.
